Fluidigm

Applications

The BioMark allows together with the Juno several applications utilizing Digital or Dynamic Array Integrated Fluidic Chip (IFC) technology. The IFC consists of networks of chambers and channels in nanolitre format, in which the PCR reactions are mixed automatically. This saves a lot of chemicals, sample material and time.

Short overview of applications:

  • High throughput gene expression analysis (with IFC): With the Dynamic Array you can do gene expression analysis of several cDNA samples (e.g. 96) against several assays (e.g. 96 in a 96.96 chip) at the same time. TaqMan and EvaGreen reagents can be used and cDNA of tissues or single cells.
  • High throughput SNP genotyping (with IFC): The Dynamic Array allows to test several samples (e.g. 96) against several SNPs (e.g. 96 in a 96.96 chip) of your choice at the same time using reagents like TaqMan probe, SYBR Green, KASP chemistry or SNPtype Assays.
  • Targeted DNA Sequencing Library Preparation System (TPS): The Juno enables the production of libraries for dozens to hundreds of samples simultaneously, with each sample enriched for up to 4,800 specific amplicons covering user-defined genes or genomic loci. Finally, the library is sequenced on a Illumina sequencing instrument.
  • Copy number variation: Using the Digital Array it is possible to distinguish the difference between four and five copies.

The following assays are available from Fluidigm but primers from any company can be loaded into the IFC:

  • SNPtype Assays: This assays are based on allele-specific PCR SNP detection chemistry. They include tagged, allele-specific PCR primers and a common reverse primer. A universal probe set is used in every reaction, producing uniform fluorescence while significantly reducing both startup and running costs.
  • DELTAgene Assays: Fluidigm designs the primers for gene expression analysis using Evagreen.

Reservation

The BioMark can be reserved via the electronic reservations system. Login with the eth login and password. Use the project number and name when entering reservations and only reserve the exact timeslots needed.
